Mitochondrial targeted meganuclease as a platform to eliminate mutant mtDNA in vivo

Abstract Diseases caused by heteroplasmic mitochondrial DNA mutations have no effective treatment or cure. In recent years, editing enzymes were tested as tools to eliminate mutant mtDNA in cells and tissues. Mitochondrial-targeted restriction endonucleases, ZFNs, TALENs been successful shifting heteroplasmy, but they all drawbacks gene therapy reagents, including: large size, heterodimeric nature, inability distinguish single base changes, low flexibility effectiveness. Here we report the adaptation of a platform based on I-CreI meganuclease known ARCUS ® . These mitochondrial-targeted meganucleases (mitoARCUS) relatively small are monomeric, can recognize sequences differing little one pair. We show development mitoARCUS specific for mouse m.5024C>T mutation mt-tRNA Ala its delivery mice intravenously using AAV9 vector. Liver skeletal muscle robust elimination with concomitant restoration levels. conclude that is potential powerful tool mtDNA.
